Pembuatan Seduhan Teh Hijau dan Teh Hitam Ditimbang teh hijau dan teh hitam sebanyak 1750 /kg, 3500 /kg dan 7000 /kg Seduhan teh dosis1750 /kg, 3500 /kg dan 7000 /kg Diseduh dengan air mendidih sebanyak 30 ml selama 3 menit lalu disaring menggunakan kain flanel 2. Pengujian metode defekasi 36 ekor mencit Diberi penandaan pada masing-masingekor mencit Ditimbang dan catat berat badanmencit Diberikan perlakuansecara per oral: suspensi CMC 1%, teh Hijau, teh Hitam dan suspensiloperamid HCl 2 30 menit kemudian diberikan Oleum ricini 0,5 ml secara p.o Diamati selama enam jam dengan interval waktu tiap 30 menit. Contoh perhitungan dosis a) Perhitungan dosis CMC Na 1% = = 10 /ml Perhitungan CMC Na 1% pada mencit 25 g = x 25 g = 0,25 ml Volume yang diberikan = 0,25 ml b) Perhitungan dosis Lodia tablet Tiap tablet Lodia mengandung 2 Loperamid HCl Dosis dewasa 2 8 /hari dan tidak lebih dari 16 /hari (berat dewasa untuk konversi 70 Kg). Dosis untuk mencit 20 g yaitu : (2 8 ) x 0,0026 = 0,0052 0,0208 /hari dan tidak lebih dari 0,04616 /Kg. Berat serbuk Lodia yang akan diambil yaitu dari 20 tablet lodia di gerus dan ditimbang = 2136. Dari 20 tablet mengandung loperamid HCl sebanyak 40. Dosis loperamid HCl yang digunakan adalah dosis maksimum yaitu : = 0,0416 /20 g mencit = 2,08 /kg Loperamid HCl yang digunakan adalah : 2,08 /kg 41

Maka serbuk loperamid yang di gunakan adalah : = x 2136 = 111,07 Dalam serbuk lodia sebanyak 111,07 mengandung loperamid HCl sebanyak 2,08. Suspensi loperamid HCl yang akan dibuat adalah 111,07 serbuk lodia dilarutkan dalam 10 ml suspensi CMC 1 %. c) Perhitungan dosis seduhan teh hijau dan teh hitam 1. Untuk dosis 1750 /kg Jumlah yang diberikan = x 25 g = 43,75 Jumlah seduhan yang diberikan = x 30 ml = 0,75 ml 2. Untuk dosis 3500 /kg Jumlah yang diberikan = x 25 g = 87,5 Jumlah seduhan yang diberikan = x 30 ml = 0,75 ml 3. Untuk dosis 7000 /kg Jumlah yang diberikan = x 25 g = 175 Jumlah seduhan yang diberikan = x 30 ml = 0,75 ml 42
