ASAM NUKLEAT METABOLISME AS. NUKLEAT Woro Anindito Sri Tunjung LAB. BIOKIMIA FAKULTAS BIOLOGI UGM 9/12/2012 1

Proses utama dlm metabolisme informasi: 1.
Pembelahan sel dan Replikasi DNA Pembelahan sel Sebelum sel membelah, sel harus membentuk dua sel structures, organelles and their genetic information 9/12/2012 22
1. Unzipping DNA - Helicase unwinds the DNA (it makes it flat, so it s not twisted anymore) - Hydrogen bonds break between the base pairs - Replication Fork = the place where e strands start t to separateate
2. Parent Strands act like a template (they act like a guide, so the matching bases know where to go) 3. DNA Polymerase (the helper ) - puts free bases on the parent strands - the bases form new strands of DNA 4. Finished! Now there are two identical pieces of DNA of DNA
* polymer = a chain of many similar pieces (DNA is a polymer. It is a chain of nucleotides.) DNA Polymerase - it creates a polymer of DNA -it proofreads the new DNA p DNA Helicase - it unwinds the double helix
The part of the DNA molecule (the gene) that the cell wants the information from to make a protein unwinds to expose the bases. Free mrna nucleotides in the nucleus base pair with one p strand of the unwound DNA molecule.
The mrna copy is made with the help of RNA polymerase. This enzyme joins up the mrna nucleotides to make a mrna strand. This mrna strand is a complementary copy of the DNA (gene) The mrna molecule leaves the nucleus via a nuclear pore into The mrna molecule leaves the nucleus via a nuclear pore into the cytoplasm

Genetic code 3 nucleotides - codon mengkode untuk 1 asam amino dlm suatu protein Codon urutan 3 nukleotida dalam mrna yang menspesifikasikan penggabungan suatu asam amino ttt mjd protein.
1. DNA unwinds PROTEIN SYNTHESIS 2. mrna copy is made of one of the DNA strands. 3. mrna copy moves out of nucleus into cytoplasm. 4. trna molecules are activated as their complementary amino acids are attached to them. 5. mrna copy attaches to the small subunit of the ribosomes in cytoplasm. 6 of the bases in the mrna are exposed in the ribosome. 6. A trna bonds complementarily with the mrna via its anticodon. 7. A second trna bonds with the next three bases of the mrna, the amino acid joins onto the amino acid of the first trna via a peptide bond. Peptide bond formation catalyzed by an enzyme complex called peptidyltransferase 8. The ribosome moves along. The first trna leaves the ribosome. 9. A third trna brings a third amino acid 10. Eventually a stop codon is reached on the mrna. The newly synthesised polypeptide leaves the ribosome.

Not all codons are used with equal frequency. There is a considerable amount of variation in the patterns of codon usage between different organisms.
Only trna fmet is accepted to form Two the initiation factors complex. (IF1 &IF3) bind to a 70S All ribosome. further charged trnas require promote fully assembled the dissociation (i.e., 70S) of 70S ribosomes into free 30S and 50S subunits. The Shine-Dalgarno sequence mrna and IF2, helpwhich ribosomes and carries mrna aligns correctly for the -GTP start of translation. - the charged trna Ribosome consists of -A bind site aminoacyl to a free 30S subunit. - Psite After these peptidyl have all - bound, E site the exit 30S initiation complex is complete.
First the mrna attaches itself to a ribosome (to the small subunit). Six bases of the mrna are exposed. A complementary trna molecule with its attached amino acid (methionine) base pairs via its anticodon UAC with the AUG on the mrna in the first position P. Another trna base pairs with the other three mrna bases in the ribosome at position A. The enzyme peptidyl transferase forms a peptide bond between the two amino acids. The first trna (without its amino acid) leaves the ribosome.
The ribosome moves along the mrna to the next codon (three bases). The second trna molecule moves into position P. Another trna molecule pairs with the mrna in position A bringing its amino acid. A growing polypeptide is formed in this way until a stop codon is reached.
Peptide bond formation catalyzed by an enzyme complex called peptidyltransferase Peptidyltransferase consists of some ribosomal proteins and the ribosomal RNA acts as a ribozyme. The process The process is repeated until a termination signal is reached.
A stop codon on the mrna is reached and this signals the ribosome to leave the mrna. A newly synthesised protein is now complete!
Termination of translation ti occurs when one of the stop codons (UAA, UAG, or UGA) appears in the A site of the ribosome. No trnas correspond to those sequences, so no trna is bound during termination. Proteins called release Proteins called release factors participate in termination
